A 4x reaction buffer developed for amplifying GC-rich DNA sequences together with TEMPase Hot Start DNA Polymerase. It contains optimised buffer components and 6 mM MgCl2, giving 1.5 mM in the reaction. It is the recommended first step when an ammonium buffer does not give acceptable results on GC-rich targets; if amplification is still poor, switch to GC Buffer II, or test both buffers side by side to save time.
