A 4x reaction buffer for amplifying GC-rich DNA sequences with TEMPase Hot Start DNA Polymerase, complementary to GC Buffer I. It contains optimised buffer components and 6 mM MgCl2, giving 1.5 mM in the reaction. It is the recommended option when GC Buffer I still does not give an acceptable result; both buffers can also be tested in parallel to speed up optimisation.
