A ready-to-use 2x master mix for amplifying GC-rich DNA sequences: it contains TEMPase Hot Start DNA Polymerase, GC Buffer I with enhancer, dNTPs and MgCl2, so only primers, template and water need to be added. The polymerase, a chemically modified Taq, stays inactive during room temperature setup and is activated by 15 minutes at 95 °C, limiting extension of mis-primed primers. It is the first choice when a standard hot start master mix fails on a GC-rich target.
