A 25 mM magnesium chloride solution in PCR-grade water for adjusting the Mg²⁺ concentration of PCR and qPCR reactions. Magnesium is essential for polymerase activity, turns dNTPs into usable substrates and stabilises the primer-template duplex: low concentrations raise fidelity but reduce activity, while high concentrations speed up amplification at the cost of fidelity and can promote primer dimers. PCR typically runs between 0.5 and 5 mM, with 1.5 mM as the starting point.
