Kit for isolating total RNA from 1-30 mg of fresh or frozen tissue, up to 25 mg of paraffin-embedded tissue, and cultured cells. The sample is homogenised and lysed with guanidinium thiocyanate and detergents, which together with beta-mercaptoethanol inactivate RNases; RNA binds to the column membrane at high chaotropic salt concentration, is washed in three steps and eluted with a low ionic strength buffer or RNase-free water. The 6-step protocol includes on-column DNase I digestion.

High binding capacity
Each column binds about 230 µg of RNA, suitable also for nucleic acid-rich tissues.
Pure RNA
A260/A280 ratio of 1.7 to 1.9, with contaminants and enzyme inhibitors removed by a three-step wash.
RNase protection
Lysis with guanidinium thiocyanate and beta-mercaptoethanol rapidly inactivates endogenous RNases.
Simple storage
Shipped at ambient temperature and stored at room temperature.
