A 5 M betaine solution in PCR-grade water, added to the reaction to improve amplification of difficult templates: GC-rich sequences, templates with strong secondary structure or otherwise problematic targets. Betaine lowers DNA melting temperature by binding preferentially to AT-rich regions, evens out the stability of AT and GC base pairs and improves the processivity of thermostable polymerases by reducing pauses caused by secondary structure. The manufacturer reports better performance than formamide, DMSO, TMAC, BSA and non-ionic detergents.
